What It Is
KPV is a tiny tripeptide (just 3 amino acids) derived from alpha-MSH, the melanocyte-stimulating hormone. Despite its size, it's a potent anti-inflammatory โ particularly for gut inflammation. It works by a completely different mechanism than melanocortin receptor agonists like melanotan. It enters cells directly through a peptide transporter and shuts down NF-ฮบB, the master inflammatory switch.
How It Works
KPV enters cells via the PepT1 transporter (no receptor binding needed) and directly inhibits NF-ฮบB activation โ the central signaling hub for inflammation. This reduces production of TNF-ฮฑ, IL-1ฮฒ, and IL-6 (the main inflammatory cytokines). It's uniquely suited for gut inflammation because PepT1 is heavily expressed in intestinal epithelial cells.
What The Research Shows
Reduced colitis severity when given orally in animal models โ one of the few peptides effective by the oral route.
Nanoparticle-loaded KPV showed enhanced efficacy for inflammatory bowel disease โ targeted delivery to inflamed gut tissue.
Reduced NF-ฮบB activation in colonocytes โ directly addresses the inflammatory driver in IBD.
Also effective for skin inflammation โ reduced contact dermatitis in topical models.
